HS036 – Restriction Mapping of pcDNA Plasmids with TRIM Gene Inserts

This experiment was to see if we could extract pcDNA3.1-TRIM6, pcDNA3.1-TRIM22, and pcDNA3.1-TRIM31 from a miniprep. The TRIMs were cut with restriction enzymes HindIII and Xbal as well as HindIII and XhoI in order to try to correctly extract the TRIM from the plasmid at the correct spot. Gel purification yielded bands at the incorrect size for TRIM6. However, TRIM22 and TRIM31 miniprep samples yielded the correct band size, therefore, they can be used for PCR.
