Phage Discovery Guide: Picking a Plaque Protocol

For POLS Labs

This video shows you how easy it is to “pick a plaque”.  You will perform this technique several times during the semester.  It will be particularly important during the phage isolation and purification portions of the curriculum.

Perform this assay only with well-isolated plaques from a plaque assay.  Do not perform this assay using plaques on a spot titer plate.  The reason for this is that it would be very easy for more than one strain/isolate of phage to be mixed together within a plaque on a spot titer plate.
